On JR line and Tokyu-Toyoko line via Shibuya is the best, I think.
From Shinjuku to Motomachi-chukagai within 1hour, 610yen but it will increase After April 1st.

Another option is Shonan Shinjuku line to Yokohama station, and Minato Mirai line to Motomachi-Chukagai. Takes about 45 min, costs 740 yen.

Or Shonan Shinjuku line to Yokohama station, and Keihin-Tohoku/Negishi Line to Ishikawacho Takes about 40 min, costs 620 yen.

In both cases, get out at the Chinatown exit.

That may depend on which station in Shinjuku you start from, but I suppose a route via Ebisu sounds simple.

If you start from a subway station on Tokyo Metro Marunouchi or Fukutoshin Line and if you can avoid morning and evening rush, also a route via Shinjuku-sanchome can be a good choice.
(Note: On weekdays in Shinjuku-sanchome Station, you cannot use the direct pathway to get from Marunouchi Line to Fukutoshin Line during 7:50 - 9:20.)

In the transfer in Shibuya, you need to get from the JR track on the 2nd floor (above the ground) to the Tokyu Tokyo Line track on the 5th basement, which was relocated in March 2013.

In the transfer in Yokohama, you need to get from the JR track on the 1st floor (= ground floor) to the Tokyu & Minatomirai track on the 5th basement.

Each of the four routes requires about 50 to 60 minutes if you start from Shinjuku Station of JR or of Tokyo Metro. The one-way fares in total from Shinjuku will be about 600 to 800 yen per adult. You can use a Suica Card (or another compatible IC card) on the whole route.

[Route via Ebisu]
Shinjuku of JR: Track 14
-- [Yamanote Linefs inner loop: 10 min. or less] --
Track 2: Ebisu: West Gate {short transfer}

Entrance gate: Ebisu (H-02) of Tokyo Metro
-- [Tokyo Metro Hibiya Line: within 5 min.] --
Track 2: (H-01) Naka-meguro {transfer at the same platform}
Naka-meguro (TY-03) of Tokyu: Track 1
-- [Tokyu Toyoko Line --(Yokohama)-- Minatomirai Railway: (#)] --
#: about 30 min. on Ltd. Express / about 35 min. on Express
Motomachi-Chukagai of Minatomirai Line: Exit 3

[Route via Shinjuku-sanchome]
Shinjuku (M-08) of Tokyo Metro: Track 2
-- [Tokyo Metro Marunouchi Line: within 2 min.] --
Track 2: (M-09) Shinjuku-sanchome
{Transfer

@rabbit2010
Although I'm not an expert and gourmet,
I love Daichin-ro and their "order viking" means " all-you-can-eat".
http://www.daichin.co.jp/tabeho.html
They say 2,380yen on weekday for adult, 2,880yen on weekend.

Today I often see "order viking" at many Chinese restaurants even in Tokyo.
