Best way from shinjuku to osaka

We are now staying at shinjuku granbell hotel. We are going to Osaka Tomorrow morning for a day tour. We just want to know what is the best and fastest way to go to Osaka from shinjuku? We are planning to use the jr station, I'm just afraid we might miss the transfers of lines. Also, what are the must see places in Osaka considering we will just stay there for the whole afternoon. Thanks.

Take a train from Shinjuku to Shinagawa or Tokyo and then take a Shinkansen to Shin-Osaka.
Please use "Hyperdia" (hyperdia.com) to check train and transfer times. :)
With almost no time in Osaka, I'd recommend Osaka Castle.
