It's possible. It takes around two hours from Tokyo.
If you have Japan Rail Pass take a shinkansen from Ueno to Utsunomiya and change to a local train to reach there.
If you don't have Japan Rail Pass the cheapest option is to take a Tobu train from Asakusa which goes straight to Nikko.

Most Tobu trains departing from Asakusa Station do not run to Tobu-nikko Station. Because there is no station beyond Tobu-nikko, if you take a Tobu train from Asakusa which is not destined to Tobu-nikko, you need to change trains at least once to get there.
On JR Nikko Line (Utsunomiya -- Nikko) only a very small number of trains are available, and you may have to wait long after getting off a Tohoku Shinkansen train.

Where and how and when in a day will you arrive? Which place in Tokyo will you get back to? Many stations are on many lines in Tokyo Metropolis. And, there are facilities like Narita Airport [NRT] outside Tokyo Metropolis which some people believe to be inside it.
