Weekends and Transportation

Hello!
I have 2 questions:
1) Is there a weekend in Japan? Meaning on some days tourist attractions/sites/places are closed or closed early? Days to pay attention to? for example Saturday or Sunday?
2) Time of transportation - How do the transportation in Japan (both Train and Buses) work at nights?

There is obviously a weekend but in general the only difference is it's much busier. Some Attractions do close but it's tends to be on a quieter weekday. Trains and buses generally finish around midnight and there are no services until around 5am.
