I recommend that you stay first either inside the HND airport, in an area around the airport or near a railroad station or bus stop which will be convenient for you to make trips.
we have 2 young kids and 2 luggages. We are looking at either staying at Asakusa or Shibuya (not confirm yet) If you arrive to Asakusa Station on Asakusa Line of Toei subway or to Asakusa Station on Ginza Line of Tokyo Metro, you may have to use one or plural staircases depending on which exit you will choose.

Shibuya has been changing; it might be a labyrinth for first-time visitors. Shibuya Stations are inside two sets of buildings, one on the ground and the other under the ground, which are located around the bottom of some slopes. A few districts surrounding Shibuya Stations have been undergoing redevelopment works. However, if none of you hates heights, you can choose one of the two hotels in Shibuya which you can reach by limousine bus.

How many travelers will be in your group including your two kids?
If you find a good candidate hotel, it is better, I suppose, for you to ask directly the hotel whether its staff is ready to have young kid guests, whether accommodation fees are required concerning your two kids and whether your kids are counted as guests regarding the capacity of rooms.
It may be not easy for you to look for, only by checking websites of hotels, a room in a medium-class hotel well-prepared to welcome young kids in Tokyo's urban area.

In the arrival lobby on the 2nd floor of the HND international terminal are bus ticket counters, which are open from 5:00 through 1:00.

Timetables for trains running to and from HND might be revised in October or November 2015.
Where else can I buy the ticket if not from the Keikyu Information Centre? There are ticket-vending machines in each of the two railroad stations of Tokyo Monorail and of Keikyu inside the HND international terminal.
While Tokyo Monorail / Keikyu trains are available from the station,
you can purchase a base ticket for a certain section at a vending machine.
Please note that you cannot reserve seats in these trains.
A Tokyo Monorail / Keikyu ride from the station inside the flight terminal requires no supplement in addition to a basic fare.
A section for which you can travel with a short-distance railroad base ticket is determined by and indicated with the station from which you travel and the amount of the basic fare. (That applies to most such cases in Japan, I suppose.)
Considering that you will not have much time to find and catch a train, I think that Tokyo Monorail will be easier for you to use as your access from HND for this time compared to Keikyu.
Keikyu trains running from HND have various destinations in plural directions including stations on lines operated by other railroad operators, while Tokyo Monorail trains are running on one simple line only.
By the way, how old will your two kids be?
Up to two young kids accompanied by one adult, when the following conditions are met, highly likely can travel by railroad in Japan free of fees.
: The kid has not reached 6 years old.
: The kid does not use a space which requires reservation in a train.
If you are not sure whether you should pay railroad fees for your kid(s),
please try, when you have enough time, to ask an attendant of the station from which he/she/they travel(s), mentioning the age of every kid.
For major railroad sections in and around Tokyo, including every section of Tokyo Monorail, of JR-East, of Keikyu, of Tokyo Metro subway and of Toei subway,
anyone of you can travel with an IC transportation card of a major brand (such as Suica and PASMO) used as a ticket, paying for basic fare(s) by the e-money stored on that card.
Please note that the price includes 500 yen paid for as a deposit. If you pay 2000 yen when you get issued a PASMO Card, for example, e-money which is worth 2000 yen (= 2000 yen - 500 yen) is initially stored on that card.
An IC transportation card can save your time to handle cash and paper tickets.
