May I know for sending yu-pack in Japan, is the box readily available at Japan Post or we need to pack ourselves?

Both ways possible.
They sell own boxes/paper bags with "yu-pack" logo.

Maybe you packed yrself is the cheapest.
Getting cardboard boxes(danbooru-bako/ダンボール箱) for free at supermarkets or convenience store.
Gummed(Gamu-tape/ガムテープ) also exists in the post office.
(100Yen shop 's one is weak, so we do not recommend it, if not light items)
