I would like to know if it possible to buy JR card (shinkanzen) from the ticket machine for return from Tokyo to Kyoto or Tokyo to Osaka.

You can buy Shinkansen tickets from the Shinkansen ticket machines at Shin-Osaka and Tokyo. From memory you can only buy one way tickets from the said origin station you are at. But unsure to be honest. Someone else should be able to confirm.

You can however buy both your to and return ticket from the ticket windows or customer service desks at the same time.

When and Where?
Basically if in a month travel, you can purchase it
at ticket vending machine and ticket office(noted open hours) both,
while in Osaka, Kyoto and any region's major JR stns
and even "KIX(airport)" or "JR Osaka" stn which has no Shinkansen tracks/rail.

But basically ticket office staff work is very faster
than vending machine(English button exist) touch screen like this

except busy time zone or someone(elder or foreigner) stucked.

But if you using "Japan Rail Pass" or other similar tourist pass,
you can not order(not purchace) at ticket machine.
(bc must show the Pass when ordering)

And, NOT Shinkan（z)en, "Shinkan(s)en" is.
Also Shinkansen ticket is not a "card" type, paper ticket like this.

(note) If use mentioned above pass, you can not use automatic ticket gate.

Shinkansen "reserved seat ticket" sales start date is just one month before fr travel date.
So if long stay travel over a month, you cannot.
(That section's Shinkansen ticket is no sale for out of Japan as online booking)

Shinkansen ticket windows and machines near Shinkansen transfer gates of Tokyo Station are set for tickets for the same-day Shinkansen rides.

At JR ticket windows outside all the JR gates of a JR station, tickets for rides on any Shinkansen sections are available.

Reservation of Shinkansen seats can be made from 10 o’clock on the day one month prior to the boarding date (#).
So, when your two Shinkansen rides are not planned within one month, you cannot buy at once your Shinkansen reserved-seat tickets for the both rides.

Among JR ticket machines, only hi-spec machines are designed to handle tickets for Shinkansen rides; those machines are installed in limited stations in smaller numbers, and their appearance, user interface and functions differ slightly by JR regional companies.

Please note that you cannot have a Shinkansen train ride only with a Shinkansen reserved-seat ticket.
To take a Shinkansen train, you both
pay a JR base fare (for the section which includes your Shinkansen section) to buy a base-fare ticket
and pay a Shinkansen surcharge (for your Shinkansen section) to buy a Shinkansen ticket.

# : In a case where the previous month does not have a day corresponding to the boarding date, Shinkansen seat reservation becomes possible on the 1st day of the boarding month.
