I would not recommend it at all. Buses tend to be small and there is little standing room, let alone room for luggage. I would place it in a station locker.

From A-B on the buses around Yokohama, I have only traveled with a small case when I was leaving/arriving for my trips. i wouldn't do it with a group or a large case. If you "had" to, pick a non-rush-hour time.

I have done this once out of necessity. It wasnt popular. Better to go to the station & put your luggage in lockers. & then bus around.
