It depends on your definition of best. @Hakata14 has given the direct route. If you are not in a hurry you could go this way. http://bit.ly/2Gx1Zqs and stop at either Onomichi or Takehara on the way.

(Yes I know I'm being a bit of a dick but I don't mean anything bad by it)

Taking a slow train along the Kure line and watching the coastline slide past ins't a bad way to waste a few hours. Trains come hourly, so you jump off somewhere you like, explore, and catch the next train.

One day with a few hours I would like to do the coastal route (only been as far as Kure so far.
A more usual route is go back to Okayama and shinkansen to Hiroshima. About 100 minutes from Uno.

I'm sure the OP was talking about the shinkansen route hence my comment above :)

@JapanCustomTours Everyone knows Onomichi but Takehara is well worth the visit for the preserved buildings and it's nicely off the main tourist route. I visited without realising that the anime Tamayura Hitotose is set there. On this website the bottom half of the page is about Takehara after you have got past the rabbits :) https://www.takeharakankou.jp/en/
