Hi
Can use JR East Pass (Nagano, Niigata) for the above Parks?
or JR Tokyo Wide Pass is good enough?

JR Tokyo Wide Pass is fine. I used it 2 years ago. Be sure you use it for travelling to other places as well, to get the value out of the pass.

As said above, yes. But please be aware that from Tokyo it's about 2 hours to get to the Hitachi Seaside Park, and with a good connection, about 90 minutes by train to get to the Ashikaga Flower Park. Then 2 hours back to Tokyo. That's pretty brutal for one day.

1. Which ways better? Tokyo to Hitachi Seaside Park then from Hitachi Park to Ashikaga Park Or Tokyo to Ashikaga then to Hitachi ?
2. If can only go 1, which has more to see in mid April?

If you want to visit both in a day, Hitachi Seaside Park and then Ashikaga Flower Park. The later open until 18:00. You need to start early, a direct LTD. EXP TOKIWA 51 departs Tokyo at 07:23. You will have limited time, possibly max two hours in each place.
