No, you don't have to. You never do.

You don't have to do anything. The real question is, to save money, "Should I buy a JR Pass?
And with your itinerary, good God, no.

Use the Rail Pass Calculator in the Transport section of this website.

It will tell you that a JR Pass will not pay off.

You are doing a one-way trip Tokyo to Osaka and a bit of other travel 15,000 yen to 20,000 yen probably. That is much less than the 7-day JRpass at 29,110 yen. Do not buy a pass.
For Pasmo/SUICA, again, you do not have to have one - for the first five years of my travels around Japan I never bothered because buying tickets was fast/easy for me. But a SUICA card has other uses and when I am looking after multiple people it is easier to use for trains, buses and in other situations.

If you really want to solve your doubts about these two transportation passes, I suggest you checking Hyperdia. Hyperdia is the number one Japanese online transportation planning tool, which gives you direct access to the most updated schedules, train information, prices to and from the selected destinations. This guide may interest you, as it's explained step by step how to use it: https://www.jrailpass.com/hyperdia
