Do I still need the basic fare using Suica before boarding the train please?

It is not a matter of calculating. It is a matter of looking it up. The base fares are set based on distance traveled. Tobu Railway has its own tables. Note that there is one table for IC cards (down to the 1 yen unit) and one for paper tickets (rounded to 10 yen). But for the ordinary traveler it is easiest to simply look them up on a site like Jorudan:

It is is 1400 yen one way from Asakusa to Nikko with a paper ticket or 1393 yen with an IC card, such as Suica.
