If I was asked ''Derek-san to yonde mo ii desu ka'', how could I answer ''calling me Derek is fine''?

I guess you want to say that they can call you "Derek," dropping the "-san," correct? Then you can say: - "Derekku" dake de ii desu yo. (Simply "Derek" would be fine.) - "Derekku" to yonde kurereba ii desu yo. (If you call me just "Derek" that would be fine.)

When calling someone's name without -san, you can use the word "yobisute", so you can say:
yobisute de iidesu
or, just say
-san wo tsukenakute iidesu (you don't need to put -san)
