'I enjoyed my work and stay in Japan'

I assume you mean "I enjoyed [working and living] in Japan" (The phrase "in Japan" describes both work and stay.) The above says "I enjoyed [working] and [living in Japan]" (The phrase "in Japan" only reaches "stay.")

So just changing the word order a bit:
- Nihon de no shigoto mo seikatsu mo tanoshikatta desu.
To make the sentence flow better, a rephrased suggestion follows:

- Nihon de seikatsu shite hataraku koto wo keiken dekite, ureshiku omotte imasu. (I am glad to have been able to experience living and working in Japan.)

whoa, these people are forgetting cultural translations, i.e., nuance

at the end be sure to say お世話になりました。 osewa ni narimashita

based on my experience any Japanese would certainly do the same

how about something like
おかげさまで日本での生活は本当に楽しかったです。みんなさんに感謝しています。お世話になりました。
okagesama de nihon de no seikatsu ha hontouni tanoshikatta desu. minna-san ni kansha shiteimasu. osewa ni narimashita
''Thanks to you (all) my life in Japan was really great. I'm grateful to you all. [can't translate -- something like you've been a big help to me, I'm indebted to you]''
