Does this mean i have to call now to confirm the date.
Or does it mean that i have to call on the date i have booked to confirm?

I am assuming that this is a comment from a hotel/inn, where you made the reservation? They are asking you to call "on the date booked" to tell them that you will certainly be coming later in the day :)
If they meant to say "now" or "today," they will say 本日 (today).
