Hello,
I will be traveling to Japan and stay in Osaka for 5 days. Please advise what transportation will be available around that time to visit Kusatsu Onsen and estimated travel time? Also, how far is Manza Onsen from Osaka as well. Thank you...j.shin

Not close, take Shinkansen or night bus until Tokyo stn or Shinjuku Bus terminal first usual. https://www.kusatsu-onsen.ne.jp/guide/en/access/If u look only at Google Maps, u may wonder why we can not go straight, bc there is no high-speed transportation like airplane and Shinkansen. Also, there is a bus route fr Karuizawa in the south of Nagano prf on the left(west), fr the northern part of Nagano City, the shortest distance is over higher mountain by taxi or car rent, so very unusual, u may have to think plan-B.

You have a long, long way to go from Osaka to Kusatsu or Manza Onsen, which may require more than six hours.
I suggest you go first to Tokyo (mainly by the Tokaido Shinkansen), stay overnight there and make a trip to Kusatsu and Manza Onsen, via Naganoharakusatsuguchi and Manza-Kazawaguchi Stations.
Kusatsu Onsen (07:00) | Bus line (07:25) Naganoharakusatsuguchi Station (07:34) | JR Agatsuma Line: Trains run very infrequently. (07:48) Manza-Kazawaguchi Station (08:32) | Bus line: Buses run very infrequently. (09:15) Manza Bus Terminal.
Have you considered flying out from Narita [NRT] or Tokyo Haneda [HND]?

Does it need to be Kusatsu onsen? I mean if you plan to go to Tokyo area anyway it makes sense. But if your trip is centered around Osaka, Kusatsu onsen is very far away. Why not some other onsen town eg in Wakayama Prefecture?eg Dorogawa onsen?
Enjoy your trip to Japan!
