Your Tobu Nikko Pass should be activated, I think,
when you get into the entrance ticket gate for the first time;
that means, you don't have to waste one day of its validity term.
The first direct train bound for Tobu-nikko is
Limited Express Revaty Kegon 1;
all the seats require reservation.
Tobu Nikko Pass does not cover the Limited Express surcharge;
you pay additionally
1470 yen (price until March the 17th, 2023) or
1650 yen (price from March the 18th, 2023)
for your ride from Asakusa to Tobu-nikko.
On Saturday, March the 18th,
Tobu Railway (and many other major railway operators)
will revise train timetables.
Regarding this Limited Express train,
the departure and arrive time seems to remain the same.

from subway Asakusa Stations of Tokyo Metro and Toei Subway.
I recommend you check well the route between your hotel and the station.
