Will be travelling thru and fro from Saitama, can we use our Jr Pass on Seibu line?

No, the Japan Rail Pass is only valid on JR lines.

Which train cards should I buy for Seibu Line? Will it be useful - and faster if I have one to go in and out from Saitama?

You can use the Pasmo or Suica farecards on Seibu. They don't save you any money, but they save the hassle of having to buy individual tickets each time.
Depending on where you are travelling, it may be possible to use an alternative JR route and thus travel for free with the Japan Rail Pass.
