Take a Shinkansen (Nozomi Super Express) from Shin-Osaka station to Nagoya station. Then transfer to Wide View Shinano. It takes about 2 hours. Costs 6,000.

Direct Train from Osaka

I think the easiest is the direct limited express Wideview Shinano from Osaka to Matsumoto with no changes of train. It takes about four hours. However, there is only one departure per day, at 8:58 a.m. (from Osaka station, not Shin Osaka). It costs 8920 yen. If you want to take a train at some other time, the easiest option is probably taking the shinkansen from Shin Osaka to Nagoya and changing to the Wideview Shinano in Nagoya. As far as I can tell, this costs a little over 10,000 yen and takes about 3 hours. You can look up trains using Hyperdia: http://www.hyperdia.com/ (click on English near the upper left of the screen)

You need to change the train from Osaka to Matsumoto in any case.
There seems to be direct bus services(both day and night). around 5000yen
