Is toei Oedo subway line at Station shinjuju-Nishiguchi E01 able to reach the bus terminal or i must stop at JR shinjuku station?

Shinjuku-nishiguchi Station (E01) of Toei subway and Shinjuku Station (M08) of Tokyo Metro subway are connected by pathways. These subway stations are located roughly to the north of the highway bus terminal.
If you get off a train at Shinjuku-nishiguchi (E01), you can get to the highway bus terminal via Exit A15 (near Shinjuku (M08) of Tokyo Metro, open 6:00 - 23:00), walking on a pathway under the ground.

If you come from Tokyo Metropolitan Government Buildings (known for observatories), you can take a train on Toei subway Oedo Line from Tochomae Station (E28) to Shinjuku Station (E27), which is located to the south of the highway bus terminal.

From Exit 7 (open 6:00 - 23:00) on the ground, you can walk about 400 meters to the highway bus terminal, turning right at the second corner then turning left to a main avenue.

I am coming from kumarae toei line.
I am taking bus to mt fuji which depart at 6am. Seem like the pathway only open 6am.
how do i walk to shinjuku bus terminal without the pathway?
All metro and subway line operates from 5am?

I would recommend that for you to go to the terminal, instead of going to the Shinjyuku Nishiguchi station (E01), go to Shinjuku Station (E27) of the Toei Oedo Line. The Nishiguchi station, located in the north in the Shinjuku area, there is a distance slightly to Express Bus Terminal.

From Shinjuku Station of Oedo Line, out into the ground from Exit 3, Go straight, take the second turn on the left.
If I were to start from Kuramae so as to be in time for the bus, it will follow the route 2 below.

By the way, did you finished the reservation for the bus?
I think the sales offices of the terminal, so start at 6:30, that it is not possible to just before you ride maybe where they buy a ticket.

Yes. I should have mentioned Exit 3, which is closer and to be open in the 5th hour.
(I assume you do not have to turn to the left; sorry if that�fs wrong.)

Please note that Toei subway Kuramae Station has two separate parts which are located a bit apart, one is for Asakusa Line and the other is for Oedo Line. (A transfer between the two lines requires walking outdoors on the ground.) The track numbers are given separately to each of the two lines. Direct trains for Shinjuku (E27) depart from Kuramae (E11) at Track 2 of Oedo Line.

I recommend checking a route to the Shinjuku highway bus terminal when you purchase your bus ticket(s).
You need to get a ticket for your 6 am bus by the previous day.
: The ticket counter on the floor above the ground (open 9:00 -20:00) handles highway bus tickets until the day before the departure date.
: The ticket counter on the floor on the ground, which handles tickets for highway buses departing on the day, is open only from 6:30 in the morning.
