Hi We'd like to visit ISAK school and town in Karuizawa, Matsumoto Castle, and snow monkey park. We have 3 nights to spare before flying out to USA. We will arrive Karuizawa by train from NRT airport around 2 PM, and will spend the 1st nt there.

Is it wise to spend the other 2 nts in Karuizawa (to avoid packing and repacking) and visiting the Matsumoto Castle and snow monkey park by trains?

No, I don't think that Karuizawa makes a good base (although not horrible either especially if you have a Japan Rail Pass or JR East Pass). Geographically a more suitable base for visiting the three places would be Nagano City which is situated in the middle of all of them. But I personally would prefer to stay in each city, assuming that I don't have a lot of luggage.
