Hello!
We are a couple, traveling to Japan on April. We will be staying in part of our trip in Tokyo, and we would like to visit Fuji Five Lakes on one of the days as a day-trip from Tokyo.
My questions is where in Fuji Five Lakes should we visit? In which location and how do we get there?
And of course - is it worthwhile? Interesting in this season?

Kawaguchi-ko is easily reached by direct bus to and from Shinjuku in Tokyo. In average years mid April is cherry blossom season there and its annual sakura festival this year is held Apr 11-19.

If you are going during the latter half of April the Fuji Skibazakura Festival near Motosu-ko is highly popular. Monitor the following links for update information on the exact date of the event this spring. During the festive event there is also direct bus going there from Shinjuku.
