Can I do the seat reservation at Shinjuku Station (Keio Highway Bus Terminal). It's site is on Japanese only and I can not understand how to do reservation

You may call them for reservation.
Keio Highway Bus Reservation Center: 03-5376-2222 (9:00`18:00) OR
Fujikyu Highway Bus Reservation Center: 0555-72-5111 (7:30`18:30)

You can do it in person at the terminal. There's a counter for the fuji route and usually a couple of good english speakers there.
I did it the morning of my climb and had no trouble getting tickets.
