Would possible to go leave Osaka, do Magome-Tsumago Trail, and go to Takayama for the night in same day? Just curious if this is all possible in the same day. Would save time and allow me to stay longer in the Kamikochi area afterwards. Thanks in advance!

Possible? Yes. Rushed? Very. Get an early start!
Instead, I'd suggest the following:
Osaka 7:49 (or Shin-Osaka 8:03)
Narai 11:25 (also known as Narai-juku)
See Narai and have lunch
Narai 13:25
Matsumoto 14:15
Possibly stay Matsumoto one night, see the castle and other sights (eg: Stay at Tabishiro Guesthouse or Matsumoto Backpackers -- MBP is right by the station and the owners of MBP are mountain climbers and very knowledgeable about the area) and then get an early start for Kamikochi. Or if you're in a huge rush, skip Matsumoto and go straight to Kamikochi.
