I have sent 2 inquiry on doing a homestay. Unfortunately, there was no reply. I sent that inquiry about 1 week ago. Does this means they are not interested to do the homestay with me??? I have not been to Japan before and their reply is very important to me as i will be travelling alone.

It sounds like they are not interested. If it is a company you are going through, I would not use them as they are not following through.
