I would ideally like to travel from Hiroshima via osaka to Koyasan, then across to ise mie, up to kanazawa followed by takayama and then kawaguchiko before reaching Tokyo. Is this realistic and feasible or am i dreaming. I dont want to spend full days doing train transfers (i have been to japan before) so is there bus connections between these places? particularily koya to ise, and from takayama to kawaguchiko? Thanks :)

Buses and local trains cost about the same. So, on routes which are well served by local trains, there often exists no bus route.

Hiroshima - Osaka can be done by bus:

The trip from Koyasan to Ise is best done by train. The easiest (and quite inexpensive) way would be taking Nankai and Kintetsu trains via Namba (Osaka).

The journey from Ise to Kanazawa leads via Nagoya. Ise to Nagoya is again best done by train:

From Nagoya to Kanazawa, the bus provides the most "cost-convenient" option (4 hours, 4,060 yen).

From July, the bus trip from Kanazawa to Takayama will be shortened from three to two hours thanks to the completion of a new expressway section. The one way fare is 3300 yen.

From Takayama to Kawaguchiko, you can take a bus from Takayama to Matsumoto (2.5 hours, 3100 yen), then a train to Kofu (2 hours, 1890 yen by local train) and then a bus from Kofu to Kawaguchiko (1 hour, 1430 yen).
