I would really appreciate your advice and answers to my questions (in italics). Is the itinerary too tight and what transportation should use? Many thanks in advance.
Day 1 Fri
Arrive in Osaka
Should I purchase the Universal Express Pass?
Universal Studios

Day 2 Sat
Can I use the JR Pass for the place below? If no, pls advise which line to use from Umeda to Namba station

The itinerary is utterly impossible. It is way too much in way too little time.

Don't forget that the Tsukiji market closes pretty early (around 7am) and it's best to go there around 5am.
Also, make sure you check their calendar to know if it's working on the day you want to visit it:

the little red dot means it's closed (as you can see it's closed most sundays)

Don't forget that the Tsukiji market closes pretty early (around 7am) and it's best to go there around 5am.

It slows down (and auctions will have ended) after around 7am, but there is still a lot to see and eat until at least noon.

The problem is you are planning to cover 2 regions, 6 cities and 2 amusement parks, within 6 days.

I plan to go Universal studios and Walt Disneyland.
After this how to move without missing the beautiful scenery, shopping and interesting culture?
Any chance for us to see red leaves in Tokyo?

Day 1 is the arrival day. You will arrive in central Osaka about 2 to 2.5 hours after the arrival of your airplane. That leaves you only with half a day in Osaka at the most.

Theme parks like Universal Studios and Disneyland are spacious and need a lot of time to visit. I recommend to allocate a full day each.

The trip from the Osaka region to the Tokyo region takes about four hours. That is almost half of a day.

Nara is a different city from Osaka. It takes one hour to get there. At least three hours to see the main sights. In total, the side trip to Nara takes at least half a day.

If you drop Nara, the Fuji Five Lakes and Kyoto, an itinerary like the following could be possible. You still would have to cut on the long lists of attractions you intend to visit on each day.

day 1: at most half a day Osaka

day 2: Universal Studios

day 3: Osaka

day 4: Osaka - Hakone - Tokyo

day 5: Tokyo

day 6: Disneyland

day 7: departure (from Tokyo's Narita Airport, I assume)
