Thinking of doing hokkaido from tokyo via shinkansen and tour other cities along the way. What are the cities that we should not miss out.

Frankly speaking, there are no "must-see" cities along the way. The most interesting cities along the way are Sendai and Morioka: http://www.japan-guide.com/list/e1102.htmlMost major attractions of the Tohoku region (the region between Tokyo and Hokkaido) are outside of the cities and require you to travel off the direct shinkansen route into the mountainous or coastal countryside.
